Objective: The aim of the study is to determine the anti-bacterial effect of PG02 leaf decoction as a mouthwash in normal subjects as compared to Chlorhexidine 0.12% mouthwash and Normal Saline Solution using a randomized controlled double blind cross-over trial

Methodology: The project consist of 2 Phases. Phase 1 will be done to determine the acceptability of different concentrations of PG01 decoction for use as gargle based on organoleptic properties. Phase 2 shall be the study proper which will be a double blinded randomized cross-over trial with 2 treatment groups.

Inclusion Criteria:

  • Adults aged 19-59
  • Those without co-morbidities like Diabetes, Respiratory/GI illness, Infections, Immunocompromised states
  • Those without gross/obvious physical deformities in the oral cavity (e.g. cleft lip, cleft palate, cavities, edentulism, etc.) as supported by Dental Clearance
  • Those who are non-smokers
  • No known allergy to food or drugs
  • Non-pregnant
Exclusion Criteria:
  • Those with dental or periodontal conditions like severe mal-alignment of teeth, orthodontic appliances, fully crowned teeth, removable partial dentures
  • Those without Dental Clearance
  • Those who underwent nasal, oral pharyngeal instrumentation for the past 6 months like intubation, nasogastric tube insertion, surgery
  • Pharmacological history that could compromise the conduct of the study will be excluded e.g. use of antibiotics, antibacterial or bacteriostatic oral hygiene preparations, steroids, supplements within the last 2 weeks
  • Those who work in areas with exposure to infections agents

PG02 leaf decoction

a.    Collection of leaves

Tender leaves of PG trees bred at the Institute of Plant Breeding in the University of the Philippines Los Banos will be harvested, and transported in air-tight containers to UP Manila for processing.

b.    Preparation of the mouthwash

The PG02 decoction mouthwash will be prepared independently by the staff of the University of the Philippines-Manila College of Pharmacy.
