The objective of the study was to determine the anti-cancer activity of Theobroma cacao seed, mucilage and pulp extracts against MCF-7 and HCT 116. Extracts were administered at concentrations of 100 ug/uL with doxorubicin as control. An MTT-base cell proliferation essay was performed to measure cytotoxic activities while cells showing decreased survival rates were subjected to a TUNEL assay to identify apoptotic cells. All MCF-7 wells treated with the extracts showed % survival rates higher than 100%. A decrease in survival rate as the concentration of the extract could yield lower survival rates. HCT 116 cells treated with 100ug/uL mucilage (60.09%) and pulp extract (86.95%) showed the lowest % survival followed by the well treated with 10ug/uL mucilage extracts had % survival rates above 100%. For both concentrations, the seed extract was significantly different from the pulp and mucilage extract. Results suggest that optimizing the concentration of the extract can lead to lower survival rates for both cell lines. TUNEL results of HCT 116 cells treated with the extract showed positive signs for apoptosis. Further fractionation and purification of the extract, the addition of a concentration-response curve and a time -course analysis could further improve the analysis done. The assay for apoptosis may also be improved by using molecular markers as opposed to morphological and physical markers for apoptosis.
