Antiangiogenic activity of Citrus maxima (Pomelo) ethanolic extracts on the chick chorioallantoic membrane (cam).

Background: Angiogenesis, the formation of new blood vessels, is important in the growth and metastasis of tumor cells. The Pomelo, Citrus maxima from the family Rutaceae, contains high levels of flavonoids which show a significant antiangiogenic effect on cancer cells by blocking the metastatic cascade. This study aimed to evaluate the antiangiogenic activity of C. maxima peel ethanolic extract using the chorioallantoic membrane (CAM) assay in the duck's (Anas platyrhynchos) embryos. Methods: Dried Citrus maxima peels extracted through maceration, Soxhlet extraction, and rotary evaporation. The crude extract was partitioned into 25%, 50%, 75%, and 100% concentrations as the experimental group. Bevacizumab (Avastin)  and 0.9% NaCI and were used as the positive and negative controls respectively. Each setup consisted of 12 eggs. Day 9 eggs were opened at the blunt side using the windowing technique. Sterile filter paper discs impregnated with the treatment groups were placed inside, directly on the CAM. The eggs were sealed with sterile parafilm and were pre-incubated for 48 hours. The antiangiogenic activity was evaluated on the 11th day of incubation by taking photographs of viable eggs and analyze with image J, a software used to measure blood vessel density and quantify blood vessel tributaries, thereby quantifying antiangiogenic activity. Data gathered was analyzed using One-Way ANOVA followed by Scheffe's test for pairwise comparison. Results: Statistical analysis revealed that Citrus maxima at 100% concentration was found to have significant effects in decreasing the number of primary blood vessels and branching points. Conclusion: Only the highest concentration of Citrus maxima ethanolic extract  (100%) has potential anti-angiogenic activity as observed in the significant decrease in the number of the blood vessel and branching points.  Keywords: Citrus maxima, Pomelo, cancer, anti-angiogenesis, angiogenesis, VEGFR
